چکیده

When 5-bromodeoxyuridine is fed to Drosophila larvae, a variety of morphogenic lesions are found in the hatching adults. Experiments were undertaken to determine whether 5-bromodeoxyuridine is incorporated into Drosophila DNA under the conditions which induce developmental modifications. Radioautographic techniques were used to examine the distribution of [aH lbromodeoxyuridine in Drosophila larvae. Label was found in nuclei of the larval tissues and the imaginal disc cells as well as the gonads. DNA was isolated from Drosophila larvae which were fed 5-bromodeoxyuridine alone or in the presence of a thymidylate inhibitor, and these samples were examined by CsC1 equilibrium density-gradient centrifugation. In the presence of 5-fluorouracil the amount of 5-bromodeoxyuridine incorporated into Drosophila DNA was increased, and this difference was reflected as a relative increase in the denser component of BUdR-DNA in CsC1 density gradients.

Morphogenic effects of halogenated thymidine analogues on Drosophila. VI. Causal analysis of bromodeoxyuridine induced growth lesions.

Variations in the treatment conditions which affect the frequency of BUdR-induced morphogenic lesions in Drosophila melanogaster have been studied. By varying the concentrations of BUdR and FU, or by changing the duration of treatment, it has been demonstrated that increased incorporation of BUdR into DNA results in a concomitant increase in morphogenic lesions. A quantitative analysis of the d...

The role of thymidylate synthetase inhibitors in bromodeoxyuridine-induced neoplasia in Drosophila.

When 5-bromodeoxyuridine (BUdR) or 5-iododeoxyuridine is administered to Drosophila larvae together with 5-fluorouracil, many of the adult flies show growth lesions in the form of extra bristles and other supernumerary wing structures. The hypothesis that the growth lesions are the result of thymidine analog incorporation in DNA and that the effects of this incorporation are increased by feedin...

Comparison of the bromodeoxyuridine-mediated sensitization effects between low-LET and high-LET ionizing radiation on DNA double-strand breaks.

The incorporation of halogenated pyrmidines such as bromo- and iodo-deoxyuridines (BrdU, IdU) into DNA as thymidine analogs enhances cellular radiosensitivity when high-linear energy transfer (LET) radiation is not used. Although it is known that high-LET ionizing radiation confers fewer biological effects resulting from halogenated pyrimidine incorporation, the exact mechanisms of reduced radi...

The nature of DNA plays a role in chromosome segregation: endoreduplication in halogen-substituted chromosomes.

AA8 Chinese hamster ovary cells were treated with halogenated nucleosides analogues of thymidine, namely CldU, 5-iodo-2'-deoxyuridine (IdU), and 5-bromo-2'-deoxyuridine (BrdU), following different experimental protocols. The purpose was to see whether incorporation of exogenous pyrimidine analogues into DNA could interfere with normal chromosome segregation. The endpoint chosen was endoreduplic...

Quantitative studies of incorporation of exogenous thymidine and 5-bromodeoxyuridine into deoxyribonucleic acid of mammalian cells in vitro.

Methods for the synthesis of bromodeoxyuridine-Br s2 and bromodeoxyuridine-H a (BUdR) are described. The incorporation of both thymidine and BUdR into the deoxyribonucleic acid (DNA) of rat bone marrow cells in vitro was markedly influenced by concentrations of thymidine and BUdR, respectively, in the incubation media. At equivalent concentrations of respective substrate, the amount of thymidin...
